You'll probably get better advice in the tech support forum (http://forums.station.sony.com/eq/forums/show.m?forum_id=1365), but in general when the PLAY button refuses to become active, it's because it's trying to update a file that is locked by Windows or another program. Usually you can fix this by rebooting and re-running the patcher before you run anything else.
